چکیده

Methods 1) 10 weeks old Fgf10 mice (50% Fgf10 expression compared to WT) in normoxic condition: Lung function and morphometric analysis. 2) BPD model: a) Fgf10 and Fgf10 mice were exposed to 85% O2 from P0-P8. Morphometric analysis and a-Actin/vWF staining were performed at P3. b) Rosa26;tet(O)Fgf10 (gain-of-function) mice were exposed to 85% O2 from P0-P8. From P9-P45 the pups were exposed to normoxia and fed either with normal food (control) or doxycycline food (experimental) to activate the transgene Fgf10. Morphometric analysis was carried out at P45. 3) Tolerance study: Rosa26;tet(O)Fgf10 and WT mice (both 10 weeks old) were exposed to doxycycline for 2 weeks. Then survival rate, histology, Ki67 and TUNEL staining were performed.

Mice deficient in the gene for cytochrome P450 (CYP)1A1 are more susceptible than wild-type to hyperoxic lung injury: evidence for protective role of CYP1A1 against oxidative stress.

Hyperoxia contributes to acute lung injury in diseases such as acute respiratory distress syndrome in adults and bronchopulmonary dysplasia in premature infants. Cytochrome P450 (CYP)1A1 has been shown to modulate hyperoxic lung injury. The mechanistic role(s) of CYP1A1 in hyperoxic lung injury in vivo is not known. In this investigation, we hypothesized that Cyp1a1(-/-) mice would be more susc...

The triterpenoid CDDO-imidazolide confers potent protection against hyperoxic acute lung injury in mice.

RATIONALE Oxygen supplementation (e.g., hyperoxia) is used to support critically ill patients with noninfectious and infectious acute lung injury (ALI); however, hyperoxia exposure can potentially further contribute to and/or perpetuate preexisting ALI. Thus, developing novel therapeutic agents to minimize the side effects of hyperoxia is essential to improve the health of patients with severe ...

The Effect of Rosa canina Extract Against Paraquat-induced Lung Injury

Background & objectives: Paraquat is a toxic herbicide that is widely used in many countries. Paraquat selectively accumulates in the lung tissue and leads to severe pulmonary injury through the oxidative and inflammatory processes. The plant Rosa canina L. has been used for long years as an herbal medicine throughout the world. Rosa canina L. is a member of Rosacea family and shows therapeut...

Genetic ablation of the Bach1 gene reduces hyperoxic lung injury in mice: role of IL-6.

Bach1 is a transcriptional repressor of the heme oxygenase (HO)-1 gene. Bach1-null (Bach1(-/-)) mice are reported to be protected from myocardial ischemia/reperfusion injury; however, the effect of Bach1 disruption on another oxidative stress model of hyperoxic lung injury has yet to be determined. To investigate the role of Bach1 in hyperoxic lung injury, Bach1(-/-) mice and wild-type (WT) mic...

Hydrogen gas reduces hyperoxic lung injury via the Nrf2 pathway in vivo.

Hyperoxic lung injury is a major concern in critically ill patients who receive high concentrations of oxygen to treat lung diseases. Successful abrogation of hyperoxic lung injury would have a huge impact on respiratory and critical care medicine. Hydrogen can be administered as a therapeutic medical gas. We recently demonstrated that inhaled hydrogen reduced transplant-induced lung injury and...
